Description The aim of this project is the discovery and characterization of biomarkers that will be used to develop innovative kits for the precocious diagnosis of pathologies with high economical impact, as tuberculosis (TB) and paratuberculosis (ParaTB) in swine, ovine, and goats. This will also lead to the improvement of our knowledge about factors influencing the diseases incidence and will also allow the implementation of effective strategies to eradicate these pathologies. For example, one could envisage the development of a genetic selection scheme based on the molecular markers for resistance to the diseases identified in the course of the project. Paratuberculosis (paraTB), whose etiological agent is /Mycobacterium avium subsp. paratuberculosis/ (MAP), is a chronic inflammation of the intestine that affects many domestic (e.g., cattle, sheep, goat) and wild species. Tuberculosis (TB), caused in animals by /Mycobacterium bovis,/ is a chronic respiratory disease characterized by the development of nodules (so-called tubercles) in the lymph-nodes of affected animals. Both pathologies are difficult to eradicate, as animals develop clinical symptoms only several years after infection and the diagnostic tools now available result really effective only during advanced stages of the diseases. For this reason, it is of paramount importance to use combined approaches to control the pathologies. These approaches should include: (i) a diagnosis based on bio-markers whose expression profile significantly changes during early phases of the infection; (ii) the selection of resistant animals based on resistance markers. The project activities will take place in Puglia and Sicilia and will be developed in 4 principal objectives. The first objective will be the collection of biological samples and epidemiological data on ParaTB and TB. This activity will allow a prior quantification of the incidence of both diseases in the Puglia and Sicilia regions, while providing the possibility to analyse in detail the risk factors that are linked to the management of the animals in selected farms. The second and third objectives will allow the identification of gene expression (such as miRNAs) and protein biomarkers suitable for the production of a diagnostic kit that will be used to precociously identify infected animals before the onset of the clinical phase, i.e., when the ELISA test normally used is not effective in the identification of the infection. The fourth objective is based on the characterization of the genetic component of the host, to identify genes and molecular markers associated with the disease resistance. This genetic information will provide the basis for the creation of a test for the identification of the most resistant animals and to set up breeding programs based on the molecular markers, along with the use of the diagnostic tools developed. Based on the know-how derived from research partners and through the development activities of industrial partners, the project will achieve products immediately available products to be used by breeders or by monitoring institutions of animal infectious diseases. The described approaches are absolutely innovative in the animal veterinary domain given that the diagnosis through the variation of genetic expression and through the proteins on easily available biological material (blood serum) is currently well established only in the human oncology field. The outcomes of the project are numerous and significant, both on cost-effective level and public health safeguard level to have genuine and guaranteed human food, mainly the dairy products.
