SVILUPPO DI MOLECOLE CAPACI DI MODULARE VIE METABOLICHE INTRACELLULARI REDOX-SENSIBILI PER LA PREVENZIONE E LA CURA DI PATOLOGIE INFETTIVE, TUMORALI, NEURODEGENERATIVE E LORO DELIVERY MEDIANTE PIATTAFORME NANOTECNOLOGICHE

Description Alterations of the redox cell metabolism and related pathways play a crucial role in the pathogenesis of several diseases (infectious and inflammatory, neoplastic, neurodegenerative) The attempts to use antioxidants in therapy have not been successful so far, because of the lack of specificity, of the poor understanding of their targets and of the targeting challenges. Based on a network including pharmaceutic companies, universities and research institutes operating in convergence areas, the aim of this proposal is to identify and characterize innovative therapeutic strategies exploiting molecules acting upon specific metabolic and redox-regulated pathways and to set up nanotechnology-based delivery systems. In particular, the proposal includes industrial and pre-competitive research activities, as summarized below: - Synthesis, research and development of molecules acting upon redox-dependent pathways involved in replication of influenza A virus, Herpes simplex 1 and 2 and inflammatory response triggered by infection. The antiviral efficacy of the molecules will be validated in pre-clinical cell studies in animal models of infection; - research and development of molecules able to interfere with redox-dependent pathways, whose activation plays a critical role in neurodegenerative processes occurring during aging. The efficacy of the molecules will be validated in pre-clinical cell studies and in animal models of neurodegeneration; - synthesis of new carnitine derivatives able to interfere with specific metabolic properties of tumor cells ensuring their survival and proliferation. The efficacy of the molecules will be validated in pre-clinical studies based in models in vitro and in vivo; - research of new redox-regulated molecular targets and identification of the mechanisms involved in their modulation in various pathological contexts, focused to the synthesis of novel molecules possibly acting in the diseases investigated in the proposal; - research and development of molecules able to inhibit bacterial growth through novel molecular mechanisms; - design, set up and evaluation of the efficacy of new systems of nanotechnology-based drug delivery, with the particular focus to innovative delivery in the ophthalmic context. Molecules with validated pre-clinical efficacy will undergo phase I and II clinical trials.
