Description The aim of the project is the study and application of new pegylated monoclonal antibody fragments and miRNA for therapeutic and/or diagnostic applications The main targets of the project are molecules involved in the neo-vascularisation processes such as the Endothelial Vascular Growth Factor (VEGF) and its Receptors (VEGF-R), the Endothelial Growth Factor (EGF) and its Receptors (EGF-R). The project will deal also with another marker potentially useful as a diagnostic indicator in chronic myeloid leukemia, the ABL/BCR out of frame protein (OOF) which is specifically expressed in these tumoral cells. For all these targets antibodies from phage libraries will be selected and conjugated with polyethylene glycol (with various molecular weights) to improve their therapeutic or diagnostics characteristics. It is known that small non coding RNA fragments (microRNA or miRNA) control gene expression both in normal and pathologic conditions. The project is going to study possible applications of miRNA-221/222 as potential inhibitor of the kit receptor expression as well as the use of these miRNA in kit dependent tumors (i.e. gastro intestinal stromal tumors (GIST) and some acute leukemia (AL). At the same time new synthetic ways for these miRNA will be investigated in order to block their intra- and extra-cellular degradation and to facilitate their penetration into cellular membranes or the target cells. This research project is therefore integrated in a very advanced sector of biomedical research with good perspectives for the development of proprietary innovative biological drugs in a sector, such as that of new anti-angiogenic drugs, of highly interesting in the oncology field. The project will allow the maintenance of a high scientific and technological concentration of high innovative potential characterized by an organic collaboration between industrial researchers and academic researchers.
