Description The project deals with the industrial development of molecules of interest in human health. The problems related to the research of such molecules are mainly the long times and high costs of tests on animals and humans in GMP (Good Manufacturing Practices). These experimental practices are necessary for the approval of the products, indeed a large part of tested molecules does not reach the approval in the reason of their toxicity. The high costs of production could be reduced if intermediate experimental protocols, providing information about toxicity of the new products before experimental practices on animals or humans, become available. Such protocols would decrease the useless experimental costs of potentially toxic molecules. In the present project, molecules with potential pharmacological or nutritional properties will be developed and tested using a biotechnological multidisciplinary platform that will give quantitative information on the potential toxicity. Among the developed substances there will be proteins of the HSP (Heat Shock Proteins) family, with known therapeutic effects and low molecular weight plant molecules. Representative human targets will constitute the platform. The interaction of molecules with the targets will be studied. Target selection is very important. Hexogen molecules exert their effects for more than 80% on cellular membrane systems such as transport proteins. After the intracellular absorption, they also interact with intracellular molecular systems. Toxic effects are caused by alterations of functions of molecular systems. Starting from these considerations and from the group expertise, molecular targets will be chosen, such as plasma-membrane transporters that mediates nutrients and cofactors absorption, endocellular systems like mitochondrial carriers and enzymes involved in bioenergetics. To optimize the target choice, genetic variability in human will be studied. These tests will select the target that have minor influence on individual variability. The variability, however, will be an important parameter to understand the function and to predict the contribute of individual genetics on the action of produced molecules. It is important to underline that both drug interactions with transports systems and pharmacogenomics are officially recognized by FDA (Food and Drug Administration) as biotechnological approaches to identify new biomarkers that could facilitate drugs development. To obtain a complete evaluation of the effects of these molecules, the interactions with cellular models and organs will be studied. Human cellular lines, isolated and perfused rat hearts and livers, will be used. These models are similar to human and already tested as experimental models. To predict modifications or effects before absorption, the interaction with intestinal microbiome will be evaluated. The potential toxicity will be evaluated studying parameters like IC50 that can describe the hexogen molecules effects on model systems. Moreover some known drugs will be tested as control of the experimental system. The data will be processed by existing software or by new software that will be designed through informatic expertise. As results, this project will supply new biologically active molecules, selected on the basis of their low toxicity that, after patent, will be introduced in future trials to be launched in the market. The biotechnological platform will be also used to supply consulting to other industries. This project pave the way to new occupational strategies both for industries and tertiary, to manage processes and data. The different research groups involved in the project proposal have a considerable know-how in the production of molecules of biological interest, and also in drug interactions with experimental systems proposed. This assures the fulfilling of the foreseen objective.
