PIATTAFORME NANOTECNOLOGICHE INNOVATIVE PER IL DELIVERY DI FARMACI IN OFTALMOLOGIA

Description The aim of this project is the study and preclinical/clinical validation of new technological platforms of drug delivery applied to the set-up of new experimental pharmaceutical formulations in the field of ophthalmology. The problem of pharmaceutical formulation in ophthalmology is particularly relevant because both the anterior and posterior segments of the eye possess anatomical, structural and functional characteristics that strongly reduce the bioavailability of the drugs administered to target tissues. This project will study highly innovative delivery systems based on nanotechnogical plattforms to target some drugs to the anterior segment and other drugs to the posterior segment of the eye. The major problem in ocular therapeutics is the attainment of an optimal drug concentration at the site of action. For the anterior segment drug delivery, common routes of administration include topical instillation and subconjuntival injection. Bioavailability of drugs from eye drops is severely impeded by a short precorneal residence time of the drug, mainly due to mechanisms of the eye that protect from foreign materials. Therapies addressed to the posterior segment of the eye are difficult too, due to very low bioavailability of the drugs on the target tissues. In fact these structures are scarcely targeted by drugs topically administered on the surfaces of the eye; consequently it is necessary to administer drugs by systemical route (with high risk of adverse effects on other structures) or to adopt invasive strategies, such as intravitreal or subconjunctival injections. To overcome the problems of conventional ocular therapy, such as short residence time, drug drainage and frequently instillation, newer drug delivery systems are being explored to improve ocular bioavailability. In general ocular bioavailability may be enhanced by increasing corneal drug penetration and by prolonging precorneal residence time of instilled drugs. Various approaches such as viscosity enhancement, use of muco-adhesive agents, hydrogels, microparticles, nanoparticles, microemulsion, lyposomes and pro-drugs have been studied. In order to solve the typical problems of the conventional formulations, in this project innovative formulations based on nanosystems will be studied to obtain an improvement of the bioavailability of the drugs on the target tissues.
