Description This project is a follow up of a previous research programme developed by S&TP of Sicily within the frame of the National Operational Programme 2000-2006 “New technologies and processes for the protection of the environment and territory and for a better management of the material life cycle”, which has generated a patent for the microbial conversion of exhausted fried oils into mcl-PHA, used to coat paper and fiber sheets successfully tested for mulching small soil plots cultivated with tomato, pepper, squash. Further research carried out by the actual proposer have allowed to discover pseudomonad bacteria and other media able to improve the performance of the elastomer, offering other and better fields of application of products and sub products of the fermentation process. The proposal aims to the valorisation of basic knowledge’s and technologies available in the sector of metabolic activities for the production of PHAs by fermentation and by heterologous systems, the study of the properties of biomass and biosurfactants, the evaluation of the applications as referred to films coated with PHA for food packaging, biomass based sheets for biodegradable and compostable bags and pots for moist organic waste and nursery plant application, antimicrobial biosurfactants. The project is structured in four main objectives: OR1-Biomass production, polymer extraction and implementation of functionalised films. Efficient systems of PHA rich biomass by the conversion of glycerol as carbon source and other waste products suitable for the biosynthesis of fat acids will be developed. An efficient process of extraction of PHA will be developed to coat paper and fiber sheets. The materials and products will be characterized in order to understand the potential application through the study of structure-function relationships. OR2- Designing and manufacturing of a prototype for the application of polymer to suitable supports. The machine is intended to uniformly apply the polymer and, if necessary, allow the polymerisation by heating process. The prototype will be tested by standardized procedures. OR3 – Molecular aspects of polyhydroxyalkanoates and biosurfactants synthesis. The goal is to develop genetic-molecular knowledge coherent to achieve a bioreactor of the suitable to reduce the cost of production by a 10 factor. In this respect the genome sequence of microbial producers and the analysis of genes involved in the biosynthesis will allow the comprehension of the regulatory systems and to transfect plants with genes responsible of PHA synthesis. The achievement of this result will allow to evaluate innovative strategies for the improvement of the production. OR4 – Field evaluation and product validation. Laboratory and field tests will be performed to evaluate the results of the application of biomass and biopolymers to obtain functionalised sheets for the production of bags for the field control of parasites, pots and bags for the nurseries, and wasting bags for the moist organic waste to compost.
