Description	The present proposal foresees the identification and multiparametric optimization of novel chemical entities, and specifically of inhibitors of Sirtuins (SIRT1, 2 and 3) and of selected histone-acetyltransferases (as phenocopiers of SIRT1 activation), to set the foundations for their further clinical development as novel pharmaco-therapies in the neurodegeneration, oncology and cardiovascular therapeutic areas. This interdisciplinary project is structured in 4 operational objectives. OBJECTIVE 1: IDENTIFICATION AND CHARACTERIZATION OF ACTIVE MOLECULES (HITS). •Activity 1 – Selection, virtual screening and acquisition of compound libraries. • Activity 2 – Assay development and primary screening, and profiling (chemico-physical properties) of active molecules. . • Activity 3 – Assay development and selectivity studies. OBJECTIVE 2– HIT OPTIMIZATION AND BIOLOGICAL VALIDATION • Activity 1 – Analogue design, synthesis and validation of identified actives. • Activity 2 – Structure-activity studies and evaluation of therapeutic potential in disease-based biological assays (neurodegeneration). • Activity 3 – Evaluation of therapeutic potential in disease-based biological assays (oncology and cardiovascular). • Activity 4 – Biophysical studies. OBJECTIVE 3 OPERATIVO 3 – LEAD OPTIMIZATION AND IN VIVO POC. • Activity 1 – ADMET profiling of compounds and further medicinal chemistry optimization. • Activity 2 – in vivo POC of lead molecules. Activity 3: identification of pharmacodynamic and predictive biomarkers. OBJECTIVE 4- PROJECT MANAGEMENT. Activity 1: Monitoring of Research Activities. Activity 2: Progress Reports • Activity 3 – Creation of a password protected web portal for dissemination of data amongst participants. • Activity 4 – Scientific dissemination (peer-reviewed publications and conference reports).
